Joey Potter, of course, is instantly intimidated, and thus would begin the infamous Jen vs. Joey Debate of  The youngest cast member at the time, Williams kicked off the first of countless tearjerker moments peppered throughout the series just from her first solo performance.Not gut-wrenching in the same way, but all the same. Jen instantly picks up on this backstage as they watch Joey perform, and Dawson grasps it even harder afterward when he and Joey have yet another conversation about how their feelings are changing. This two-parter revolved around Jack McPhee — who is new to Capeside, alongside his sister Andie — coming to terms with the fact that he is gay and being forced by the actual worst teacher in the history of teaching to read a poem out loud that he is not ready to read.And he and his dad do work it out — but not for a season and a half. It would create the network brand of beautiful angsty teenagers maybe having sex in beautiful nostalgic Americana landscapes.It was the new show that everyone had to talk about. Which meant, for one of the first times in TV history, it was the new show that everyone had to talk about on the internet. Sarah D. We read these recaps, which were called wrap-ups, by Danny Drennan. And we then just started chatting offline. But over time, I sort of fell in love with the fact that they had this kind of heightened language that was their own, but their emotions and all of the things that they were going through still made them very much teenagers.That tension, you know, I really liked. They just all sounded like characters on a TV show. They were very hyperverbal. That became the charm of the show and what people liked about it; that it was this heightened reality.That was annoying to me at the time, but ultimately, Dawson was a problem. Dawson was just such a pill. And he had been so good to me and had really kick-started my entire career.So I did it. Possibly so that we would shut the fuck up and let them go back to talking like normal people. But possibly because they thought it would be a good idea, which it turned out to be. It did well enough in its first year that, after that, we went back and added a bunch of other shows. And Dawson was our flagship show. This must be nice!And it also seemed to be oblivious to what it actually did do well. I give it credit: It learned. It developed an intelligence about itself. And crying Dawson is Exhibit A. The TV show that the WB had built its brand around just two years earlier was now in danger of cancellation.So I was the only remaining writer from previous seasons that was on the staff at the top of season three. I assumed this was just TV. I went off to make a movie, and I came back, and the network was making some changes to the folks that were above me, who I quite liked. And then we also hit a wall with this storyline where Pacey and Jen were going to embark on a casual fuck-buddy relationship.I guess somewhere around episode eight, the network flipped out. Especially that season. We had a production shutdown around Thanksgiving. Bunting, cofounder of Television Without Pity: It seemed as though, perhaps because Kevin Williamson had left, the rest of the team maybe felt a little bit freer to let the show become what it wanted to become. Which was not a show about Dawson. But the heart wants what it wants, and that can complicate things.Tara Ariano, cofounder of Television Without Pity: Pacey was obviously the better character and always had been. And the two of those actors were like magic to watch.I said I would do it if they let me have an actual gay kiss between two characters. That had never happened on network TV before. And the network agreed to that. So after that, I felt like it was worth it. And also we need to end a lot of the other storylines and stuff. But it was really connecting with the audience. But the ratings started to grow again. The actors were happy, and everybody was excited. I think we found what the show would be without Kevin there.       Sequencing machines can't "see" DNA directly, so scientists must use a complex set of procedures to prepare DNA for sequencing. When DNA is finally in a form that the machines can read, it has been chopped up, copied, chemically modified, and tagged with fluorescent dyes corresponding to the four different DNA bases, or genetic letters. Before it is sequenced, a piece of DNA is copied many times, then divided into four batches in preparation for another round of copying.When one of these modified bases is incorporated into a DNA molecule, the chain of bases stops growing. The result of all this is that one batch of DNA will contain only pieces that end in T, another only pieces that end in A, a third only pieces that end in G, and the fourth batch only pieces that end in C.In the second round of copying, a different fluorescent dye is also added to each batch of DNA. Thus, every piece of DNA that ends with T has a blue dye tag, for example; those that end in A have a red dye tag; those that end in G have a yellow dye tag; and those that end in C have a green dye tag.At the end of the second round of copying, each batch will contain the following pieces of DNA:. Into one lane or capillary of a sequencing machine goes a mixture of DNA from all four batches.Thus, in this example, the first piece to make it all the way through the gel is a T attached to a blue dye tag; the next piece is TA with a red dye tag; next is TAG attached to a yellow dye tag; and so on. As the pieces emerge from the gel, they move past a laser that causes the dye molecules to fluoresce. Thus, although it was met with skepticism at its inception, the Human Genome Project will certainly be heralded as one of the most important scientific endeavors of our time.Within a span of only 13 years, an amalgam of public and private researchers was able to successfully complete the Human Genome Project. Although these scientists used a number of different methods in their work, they nonetheless obtained the same results. In doing so, the researchers not only silenced their critics, but they also beat their own estimated project timeline by two entire years.Perhaps even more importantly, these scientists inspired an ongoing revolution in our fight against human disease and provided a new vision of the future of medicine-although that future has yet to be fully realized.
